Vi-Typhoid Conjugate Vaccine (Vi-TCV) is a typhoid fever vaccine that consists of the purified Vi capsular polysaccharide from Salmonella enterica serovar Typhi chemically conjugated to a carrier protein such as tetanus toxoid (TT) or diphtheria toxoid (DT). This conjugation enhances immunogenicity and allows for effective vaccination in children as young as six months old. The mechanism of action involves stimulating the immune system to produce anti-Vi antibodies that protect against S. typhi infection. Unlike older polysaccharide vaccines, which are not effective in children under two years and require multiple doses or boosters for sustained protection, TCVs provide strong protection with a single dose and are suitable for routine childhood immunization programs. WHO recommends TCVs due to their improved efficacy and safety profile; they have been prequalified by WHO and introduced into national immunization programs in several countries[4][5][8].
