Viagenpumatucel-L is an allogeneic, cell-based therapeutic cancer vaccine developed for the treatment of advanced non-small cell lung cancer (NSCLC). It consists of a human lung adenocarcinoma cell line engineered to express multiple tumor-associated antigens (including MAGE-A3, NY-ESO-1, LAGE-1, and others) chaperoned by a modified secretory heat shock protein gp96 fused to immunoglobulin (gp96-Ig). This fusion protein acts both as an antigen delivery vehicle and immune adjuvant. The vaccine delivers these antigens directly to the patient's antigen-presenting cells via the cross-presentation pathway, leading to exclusive activation of CD8+ cytotoxic T cells. This mechanism enables broad immune targeting across diverse tumor antigens and MHC barriers. Viagenpumatucel-L was investigated in combination with other therapies such as nivolumab in clinical trials for NSCLC but development has been discontinued[3][4][5][7].
