VIB585 is an investigational masked bispecific T-cell engager (TCE) developed by Vibrant for the treatment of prostate cancer. The drug is engineered using Vibrant's proprietary masking technology, which incorporates a peptide mask linked to the CD3-binding domain via a tumor-protease-sensitive linker. This design ensures that the T-cell engager remains largely inactive in the blood, potentially mitigating systemic toxicities such as cytokine release syndrome (CRS). Once VIB585 enters the tumor microenvironment, local proteases cleave the linker, removing the mask and enabling the bispecific antibody to bridge PSMA-expressing prostate cancer cells with CD3-positive T cells. This interaction triggers targeted T-cell mediated cytotoxicity against the tumor cells. VIB585 is currently in preclinical development.
