Overview

Vibepolimab (HL-063) is a recombinant chimeric (human-mouse) monoclonal antibody targeting the epidermal growth factor receptor (EGFR). By binding to the extracellular domain of EGFR, vibepolimab competitively inhibits the binding of epidermal growth factor (EGF) and other ligands, thereby blocking the activation of the receptor's tyrosine kinase activity and subsequent downstream signaling pathways, including the MAPK, PI3K/Akt, and JAK/STAT pathways. This inhibition leads to suppressed cell proliferation, induction of apoptosis, and reduced angiogenesis and metastasis in EGFR-overexpressing tumors. Vibepolimab is primarily being developed in China for the treatment of solid tumors, such as esophageal squamous cell carcinoma (ESCC), often in combination with immune checkpoint inhibitors like pucilizumab and radiotherapy.
