Vibosameran is a synthetic messenger RNA (mRNA) encoding the melanoma-associated antigen A3 (MAGE-A3). Developed by BioNTech, it is one of the four active mRNA components of the BNT111 (FixVac) cancer vaccine, which is designed to treat advanced melanoma by targeting a specific set of tumor-associated antigens. Vibosameran is formulated within RNA-lipoplexes (RNA-LPX) to facilitate targeted delivery to lymphoid tissues and uptake by antigen-presenting cells, such as dendritic cells. Once internalized, the mRNA is translated into the MAGE-A3 protein, which is then processed and presented on the cell surface to stimulate a robust CD8+ and CD4+ T-cell mediated immune response against MAGE-A3-expressing tumor cells. It is currently being evaluated in Phase 2 clinical trials, primarily in combination with immune checkpoint inhibitors like cemiplimab for patients with anti-PD-1-refractory melanoma.
