This is a combination of three anticancer drugs: **vibostolimab** (an anti-TIGIT immune checkpoint monoclonal antibody), **pembrolizumab** (a programmed death receptor-1, PD-1, blocking monoclonal antibody also known as Keytruda), and **enfortumab vedotin** (an antibody-drug conjugate targeting Nectin-4, also known as Padcev, which delivers the microtubule-disrupting payload monomethyl auristatin E to cancer cells). - Vibostolimab is designed to inhibit TIGIT, a protein that suppresses T cell activity, enhancing the immune system's ability to attack tumors. - Pembrolizumab binds to PD-1 receptors on T cells and prevents them from being inactivated by cancer cells, restoring the anti-tumor immune response. - Enfortumab vedotin targets Nectin-4 on urothelial and other cancers; it is internalized, releasing MMAE to disrupt microtubules and trigger cancer cell death. The combination is being developed for advanced or metastatic **urothelial carcinoma**, especially in patients not eligible for cisplatin-based chemotherapy. Each agent has demonstrated efficacy in various malignancies, but this specific combination is an investigational regimen under clinical evaluation.
