Overview

vic-trastuzumab duocarmazine + paclitaxel is a combination therapy under investigation for the treatment of HER2-positive cancers, particularly metastatic breast cancer. Vic-trastuzumab duocarmazine (SYD985) is an antibody-drug conjugate (ADC) that consists of the monoclonal antibody trastuzumab linked to a cleavable cytotoxic payload, valine-citrulline-seco-DUocarmycin-hydroxyBenzamide-Azaindole (vc-seco-DUBA). The ADC binds to HER2 on tumor cells and, upon internalization and proteolytic cleavage, releases an active DNA-damaging agent that induces tumor cell death. Paclitaxel is a taxane-class small molecule chemotherapeutic that stabilizes microtubules by binding to β-tubulin, preventing their depolymerization and thereby inhibiting mitosis and inducing apoptosis in cancer cells. This combination aims to leverage both targeted delivery of cytotoxin via the ADC and broad antimitotic activity from paclitaxel[1][3][5][6].
