Overview

Vidofludimus calcium is an orally administered, small molecule drug under investigation for the treatment of multiple sclerosis (MS), including relapsing-remitting, secondary progressive, and primary progressive forms. It acts as a first-in-class activator of nuclear receptor-related 1 (Nurr1), providing direct neuroprotective effects by reducing neuronal loss and injury. Additionally, it selectively inhibits dihydroorotate dehydrogenase (DHODH), leading to anti-inflammatory and anti-viral effects by targeting hyperactive immune cells without suppressing normal immune function. This dual mechanism results in reduced T-cell and B-cell activity, decreased proinflammatory cytokine release, suppression of new MRI lesions in MS patients, and potential protection against viral infections such as EBV. Vidofludimus calcium has shown a favorable safety profile with low discontinuation rates in clinical trials[1][2][3][4][5][6][8].
