Vidupiprant

Vidupiprant is a small molecule drug that functions as a dual antagonist of prostaglandin D2 receptor 1 (DP1) and prostaglandin D2 receptor 2 (DP2, also known as CRTH2). It also acts as an allosteric inhibitor of calpain-1. Initially developed by Amgen, Vidupiprant was investigated for the treatment of asthma, but its development for this indication was discontinued due to unpromising clinical outcomes. Preclinical studies have demonstrated its potential in dampening lupus-like nephritis activity by inhibiting basophil activation and recruitment. Furthermore, its calpain-1 inhibition mechanism suggests potential therapeutic applications in cardiovascular, neurodegenerative, and ischaemic diseases.
