The vimentin vaccine (specifically IMMU-23) is a conjugate cancer vaccine designed to elicit a humoral immune response against extracellular vimentin. Vimentin is an intermediate filament protein that is typically intracellular but becomes overexpressed and secreted by tumor endothelial cells into the tumor microenvironment. Extracellular vimentin has been shown to possess pro-angiogenic and immunosuppressive properties, facilitating tumor growth and progression. By inducing high titers of anti-vimentin antibodies, the vaccine aims to neutralize these effects, leading to inhibited tumor growth, increased tumor cell necrosis and apoptosis, and enhanced infiltration of T cells and Fcγ receptor-positive immune cells. Developed by CimCure BV, the vaccine has demonstrated efficacy in preclinical models of glioblastoma, improving survival and reshaping the tumor microenvironment.
