Overview

Vinblastine + nilotinib is an investigational combination therapy evaluated primarily for pediatric, adolescent, and young adult patients with refractory or recurrent low-grade glioma. Vinblastine is a vinca alkaloid chemotherapy agent that inhibits mitosis by binding to tubulin and disrupting microtubule formation, leading to cell cycle arrest at metaphase and subsequent cell death. Nilotinib is a small molecule tyrosine kinase inhibitor that targets BCR-ABL, c-Kit (KIT), platelet-derived growth factor receptors (PDGFRA/PDGFRB), and other kinases involved in tumor growth, angiogenesis, and immunomodulation. The rationale for combining these agents lies in their potential synergistic effects on inhibiting tumor proliferation and angiogenesis through distinct but complementary mechanisms of action. This combination has been studied in phase I/II clinical trials under the name VINILO[1][3][4][6].
