Vinblastine N-oxide is a **small molecule prodrug of vinblastine** developed to enhance selective cytotoxicity in hypoxic tumor environments, such as those found in non-small cell lung cancer and potentially pancreatic cancer. It incorporates an N-oxide functional group that is preferentially reduced by endogenous reductases present in hypoxic tissues, releasing active vinblastine locally and minimizing systemic toxicity. Vinblastine N-oxide (also known as CPD100) has demonstrated improved safety and toxicity profiles compared to vinblastine in preclinical models, but a shorter plasma half-life. For improved delivery and circulation time, liposomal formulations (notably CPD100Li) have been explored. The compound is structurally related to vinblastine, a vinca alkaloid class chemotherapeutic, but is not approved for clinical use and remains in preclinical development for cancer therapy[1][5][7].
