Vinburnine is a vinca alkaloid derivative, specifically the oxidized form of vincamine, traditionally utilized as a vasodilator for the treatment of cerebrovascular disorders and cognitive impairment. In recent oncological research, vinburnine has demonstrated potential as an antineoplastic agent in melanoma. Its mechanism involves the induction of reactive oxygen species (ROS), which leads to DNA damage and triggers the apoptotic cascade. Furthermore, it activates the P38/MAPK/ATF3 signaling axis, resulting in the secretion of interleukin-24 (IL-24). This process enhances CD8+ T cell activity and modulates the tumor immune microenvironment, showing synergistic potential when combined with anti-PD-1 immunotherapy.
