The LMB (Lymphome Malin B) regimen is a multi-agent cytotoxic chemotherapy protocol developed by Gustave Roussy for the treatment of pediatric high-risk B-cell Non-Hodgkin Lymphoma (B-NHL) and B-cell Acute Leukemia (B-AL). This small molecule combination regimen consists of vincristine (a microtubule inhibitor), cyclophosphamide (an alkylating agent), high-dose methotrexate (a folate antagonist), doxorubicin (an anthracycline topoisomerase II inhibitor), and cytarabine (a nucleoside analog antimetabolite). The protocol is typically administered in intensive phases, including induction and consolidation courses (such as CYM and CYVE), and often incorporates intrathecal therapy for central nervous system prophylaxis. It has served as the standard-of-care backbone in major international clinical trials, such as the Inter-B-NHL Ritux 2010 trial, which investigated the efficacy of adding rituximab to this chemotherapy foundation in children and adolescents with advanced-stage CD20-positive B-cell malignancies.
