Drug intelligence / Profile preview
Vincristine sulfate liposome is a nanoparticle-encapsulated formulation of the vinca alkaloid vincristine sulfate. By utilizing a sphingomyelin and cholesterol-based liposome, the drug achieves prolonged circulation and enhanced penetration into tumor tissues via the enhanced permeability and retention (EPR) effect. This formulation allows for the delivery of higher dose intensities compared to conventional vincristine sulfate while potentially mitigating dose-limiting peripheral neuropathy. Its mechanism of action involves binding to tubulin, which inhibits microtubule assembly and leads to mitotic arrest and apoptosis. It was originally developed by Talon Therapeutics (later acquired by Spectrum Pharmaceuticals, which was subsequently acquired by Assertio) and received FDA accelerated approval for the treatment of adult patients with Philadelphia chromosome-negative acute lymphoblastic leukemia in second or greater relapse.
