Drug intelligence / Profile preview
**Vincristine sulfate liposome injection** is a liposomal formulation of the vinca-alkaloid cytotoxic drug vincristine sulfate. Marketed as **Marqibo**, it encapsulates vincristine in sphingomyelin/cholesterol liposomes originally developed using Inex Pharmaceuticals' sphingosomal delivery technology. Vincristine binds tubulin and disrupts microtubule dynamics, causing mitotic arrest and apoptotic cell death. The liposomal carrier was designed to prolong systemic circulation, increase delivery to tumors and reticuloendothelial tissues, and slowly release vincristine, permitting higher uncapped vincristine exposure than conventional vincristine. In a pivotal Phase 2 study in multiply relapsed or refractory aggressive non-Hodgkin lymphoma, single-agent Marqibo produced a 25% overall response rate but was not granted approval for that indication. Marqibo was later granted accelerated US approval for relapsed Philadelphia chromosome-negative acute lymphoblastic leukemia in 2012; FDA withdrew that approval in 2022 after confirmatory clinical benefit was not verified.
