Vincristine sulfate liposome injection is a formulation of the microtubule-targeting agent vincristine encapsulated in sphingomyelin/cholesterol-based liposomes, designed to optimize pharmacokinetics and improve tumor drug exposure by providing prolonged circulation and sustained release. It is FDA approved for adult patients with Philadelphia chromosome-negative acute lymphoblastic leukemia in second or greater relapse or whose disease has progressed after two or more antileukemia therapies. Rituximab is a monoclonal antibody targeting CD20 on B cells, used primarily for B-cell malignancies. The combination of vincristine sulfate liposome injection and rituximab has been studied in relapsed/refractory diffuse large B-cell lymphoma, mantle cell lymphoma, and indolent B-cell lymphomas[1][2][5]. Vincristine acts by inhibiting microtubule formation during mitosis, leading to cell cycle arrest and apoptosis[6]. Rituximab induces cell death through complement-dependent cytotoxicity, antibody-dependent cellular cytotoxicity, and direct apoptosis via CD20 binding.
