Overview

Vinorelbine + docetaxel is a combination chemotherapy regimen used primarily in the treatment of metastatic non-small cell lung cancer (NSCLC) and metastatic breast cancer. This combination leverages two anti-mitotic agents with complementary mechanisms targeting microtubules to inhibit cancer cell division. - **Vinorelbine** is a semi-synthetic vinca alkaloid that acts as a mitotic spindle poison by binding to tubulin subunits, inhibiting microtubule polymerization, and arresting cells in metaphase during mitosis. - **Docetaxel** is a taxane that promotes abnormal polymerization of tubulin into stable microtubule bundles by binding specifically to the β-tubulin subunit, thereby inhibiting microtubule depolymerization and disrupting mitosis. The combination aims to exploit their synergistic effects on microtubules with non-overlapping toxicity profiles. Clinical studies have evaluated this regimen as frontline or second-line therapy for incurable NSCLC but found no significant efficacy advantage over platinum-based doublets; it has also been studied in metastatic breast cancer[1][2][3].
