Overview

Vinorelbine + ifosfamide is an investigational combination chemotherapy regimen used in the treatment of advanced cancers, particularly non-small-cell lung cancer. Vinorelbine is a semi-synthetic vinca alkaloid that disrupts microtubule formation, inhibiting mitosis by blocking tubulin polymerization. Ifosfamide is an oxazaphosphorine alkylating agent and a prodrug that must be activated in the liver to form metabolites which induce DNA cross-linking, leading to apoptotic cell death. This combination has demonstrated activity in clinical trials, with the primary dose-limiting toxicity being myelosuppression, especially neutropenia. It is sometimes administered with supportive agents like G-CSF to manage cytopenias[1][3][4][5].
