VIO-01 is a next-generation pan-DNA damage response (DDR) DNA decoy therapeutic developed by Valerio Therapeutics. It is designed to abrogate multiple DNA repair pathways, including homologous recombination and non-homologous end joining, by acting as a decoy for key DDR proteins such as PARP1, Ku70/80, and the MRN complex (MRE11-RAD50-NBS1). This mechanism disrupts cancer cell DNA repair processes and triggers a robust immune response through activation of the STING pathway. The drug consists of double-stranded oligonucleotides linked with a cellular uptake facilitator for enhanced tumor targeting and stability. It is administered intravenously and is being investigated primarily for advanced solid tumors with homologous recombination deficiency (HRD+) or HRR mutations, including ovarian cancer, breast cancer (BRCA1/2 mutant), and prostate cancer[1][2][3][4][6].
