VIP126 is a **small-molecule camptothecin payload** developed by Vincerx Pharma as the cytotoxic warhead of the small molecule-drug conjugate VIP236. It is a modified **7-ethyl camptothecin** designed to inhibit **topoisomerase 1**, thereby inducing DNA damage and tumor cell death. Relative to SN38, VIP126 was engineered for **high permeability** and **low efflux**, and is reported not to be a substrate of the multidrug transporters **P-glycoprotein** and **BCRP**, properties intended to improve delivery and activity in resistant solid tumors. VIP126 is not being developed as a standalone administered medicine; instead, it is designed to be released in the tumor microenvironment from VIP236 after **neutrophil elastase-mediated cleavage**, with development focused on solid tumors including breast, colorectal, small cell lung, gastric, and renal cancers.
