Overview

VIP716 is a highly potent and selective small molecule inhibitor of kinesin spindle protein (KSP, also known as KIF11 or Eg5), developed by Vincerx Pharma. It is specifically engineered as a cytotoxic payload for antibody-drug conjugates (ADCs). The molecule incorporates a hydrophilic CellTrapper™ moiety designed to reduce membrane permeability, which promotes intracellular accumulation within tumor cells following release and minimizes bystander penetration into healthy tissues, thereby reducing systemic toxicity. VIP716 is released from its parent ADCs via legumain-mediated cleavage. It is the active payload in clinical-stage candidates such as VIP943 (targeting CD123) and VIP924 (targeting CXCR5), which are being developed for hematologic malignancies including acute myeloid leukemia (AML) and B-cell lymphomas.
