VIP924 is a first-in-class antibody-drug conjugate (ADC) developed by Vincerx Pharma for the treatment of B-cell malignancies. It consists of an anti-CXCR5 humanized monoclonal antibody linked via a novel legumain-cleavable peptide linker to a highly potent and selective kinesin spindle protein inhibitor (KSPi) payload, known as VIP716. The linker is specifically cleaved by legumain, a tumor-associated lysosomal protease, resulting in the release of the active KSPi payload inside CXCR5-positive tumor cells. The KSPi has been modified with CellTrapper technology to enhance intracellular accumulation and reduce off-target toxicity by limiting membrane permeability. Preclinical studies have demonstrated significant efficacy in lymphoma models, including mantle cell lymphoma, follicular lymphoma, diffuse large B-cell lymphoma (DLBCL), and chronic lymphocytic leukemia. VIP924 shows superior activity compared to other ADCs targeting B-cell malignancies and may be used as monotherapy or in combination with other targeted agents[1][2][4][5][6][7].
