VIPER-101 is a gene-edited, autologous chimeric antigen receptor (CAR) T-cell therapy developed for the treatment of relapsed or refractory T-cell lymphoma. It is engineered to target CD5, a surface glycoprotein present on cancer cells in over 85% of patients with T-cell lymphoma. The therapy utilizes Vittoria Biotherapeutics' proprietary Senza5 platform, which integrates CD5 gene-editing technology and a rapid five-day manufacturing process to enhance cell potency and stemness. By knocking out CD5 in the CAR-T cells, VIPER-101 circumvents fratricide (self-killing among therapeutic T-cells) and abrogates immunosuppressive signaling mediated by CD5, thereby improving anti-tumor efficacy and safety compared to conventional CAR-T therapies targeting T-cell malignancies. The drug is currently being evaluated in Phase 1 clinical trials for multiple subtypes of T-cell lymphoma[1][2][3][4][6][7].
