VIR-5500 is an investigational, dual-masked T-cell engager (TCE) developed for the treatment of solid tumors, specifically targeting prostate-specific membrane antigen (PSMA) in metastatic castration-resistant prostate cancer (mCRPC). The drug utilizes PRO-XTEN masking technology, which keeps the TCE inactive until it reaches the tumor microenvironment. There, tumor-specific proteases cleave off the mask and activate the TCE, enabling targeted destruction of cancer cells by directing T-cells to attack PSMA-expressing tumor cells. This approach aims to reduce systemic toxicity and improve tolerability compared to traditional TCEs. Early phase 1 clinical data indicate encouraging safety and efficacy profiles with no requirement for corticosteroid or anti-interleukin-6 premedication and no grade ≥3 cytokine release syndrome events observed[1][2][5].
