VIR-5525 + pembrolizumab is a combination investigational therapy for solid tumors expressing EGFR, currently in Phase 1 clinical trials. VIR-5525 is a dual-masked T-cell engager (TCE) that utilizes PRO-XTEN masking technology to selectively activate within the tumor microenvironment, aiming to mitigate damage to healthy cells and reduce toxicity. It targets the epidermal growth factor receptor (EGFR) to direct T-cell-mediated killing of cancer cells. Pembrolizumab is a monoclonal antibody and immune checkpoint inhibitor that blocks PD-1, preventing tumor cells from evading immune attack and enhancing T-cell response. This combination is intended to leverage both direct T-cell targeting of EGFR-positive tumors (via VIR-5525) and immune checkpoint blockade (via pembrolizumab), potentially improving anti-tumor efficacy in advanced, locally advanced, or metastatic solid malignancies with EGFR expression.
