VIR-7229 is an investigational, next-generation monoclonal antibody developed for the prophylaxis of COVID-19. It was discovered using Vir Biotechnology’s proprietary monoclonal antibody discovery platform and incorporates Xencor’s Xtend technology to extend half-life. The antibody has been affinity-matured using machine learning to enhance its potency, breadth, and resistance to viral escape. Mechanistically, VIR-7229 targets the receptor-binding motif (RBM) of the SARS-CoV-2 spike protein with high cross-reactivity across all sarbecovirus clades—including non-ACE2-utilizing bat sarbecoviruses—and potently neutralizes a wide range of SARS-CoV-2 variants (including EG.5, BA.2.86, JN.1). Its high binding affinity and molecular mimicry confer a high barrier to resistance; escape mutants are rare and associated with reduced viral fitness[4][5][8]. Preclinical data show broad antiviral activity by competing with ACE2 for spike protein binding and inducing S1 shedding[3]. The development is supported in part by BARDA/HHS/ASPR funding as part of Project NextGen[6][10].
