VirDUB is a preclinical small-molecule inhibitor developed by the Walter and Eliza Hall Institute (WEHI) in collaboration with Roche. It specifically targets the papain-like protease (PLPro) of the SARS-CoV-2 virus. PLPro is a critical enzyme for viral replication and also plays a role in evading the host's immune response by acting as a deubiquitinating (DUB) enzyme, removing ubiquitin and ISG15 from host proteins. By inhibiting PLPro, VirDUB aims to block viral replication and restore the host's innate immune signaling. The drug is being investigated for the treatment of COVID-19 and potentially Long COVID.
