Overview

Viscosalactone B is a natural withanolide isolated from the plant *Withania somnifera* (Ashwagandha). It functions as a selective inhibitor of Lysine-specific demethylase 1 (LSD1), an epigenetic enzyme that is frequently overexpressed in various malignancies, including prostate cancer. By inhibiting LSD1, viscosalactone B induces the accumulation of methylated histone substrates such as H3K4me1, H3K9me1, and H3K9me2, which subsequently alters gene expression to suppress tumor cell proliferation. Preclinical research has demonstrated its antiproliferative activity against multiple prostate cancer cell lines, including those resistant to the second-generation anti-androgen enzalutamide (MDV3100). In vivo studies in xenograft models have shown significant antitumor efficacy with minimal systemic toxicity, suggesting its potential as a therapeutic candidate for prostate cancer treatment.
