Drug intelligence / Profile preview
Visfatin A-domain peptide is a synthetic peptide corresponding to the active site domain (amino acid residues 181 to 390) of visfatin (also known as nicotinamide phosphoribosyltransferase, NAMPT). Visfatin is a 52 kDa adipokine with known angiogenic and cell-proliferative activities, but its large molecular weight limits its direct therapeutic development. The A-domain peptide was designed to overcome these limitations by retaining the active site responsible for visfatin's biological effects. In preclinical models of 4-vinylcyclohexene diepoxide (VCD)-induced ovarian failure, administration of the visfatin A-domain peptide significantly improved ovarian function and fertility potential. It acts by stimulating ovarian angiogenesis and early follicular development, as evidenced by increased numbers of primordial and primary follicles, elevated serum anti-Müllerian hormone (AMH) levels, and upregulated expression of angiogenic and follicular development factors such as BMP15, C-KIT, and VEGF. The peptide is being investigated as a potential therapeutic strategy for ovarian aging and female infertility.
