Overview

VISK-103 is an antibody-enzyme fusion protein developed by Viska Bio for the treatment of glioblastoma (GBM). It consists of an EGFR-targeting nanobody fused to a modified xanthine oxidase (XO) enzyme domain. The drug employs a three-pronged mechanism of action: the nanobody selectively binds to and inhibits EGFR-positive tumor cells; the XO domain generates extracellular superoxide to induce cell death in both target and surrounding cells; and the resulting immunogenic cell death (ICD) triggers the release of damage-associated molecular patterns (DAMPs), recruiting and activating immune cells within the immunosuppressive tumor microenvironment. Preclinical studies in GBM xenograft models have demonstrated significant reductions in tumor burden and improved survival.
