VISTA CAR-T is an experimental second-generation chimeric antigen receptor (CAR) T-cell therapy designed to target the V-domain Ig-Suppressor of T cell Activation (VISTA). VISTA is an immune checkpoint molecule predominantly expressed on myeloid cells and often upregulated by acute myeloid leukemia (AML) cells to evade immune surveillance. The CAR construct consists of an scFv derived from an anti-human VISTA antibody, a CD8α hinge/transmembrane domain, a 4-1BB costimulatory domain, and a CD3ζ signaling domain. Developed by researchers at the University of Minnesota and Vanderbilt University, this therapy has demonstrated potent in vitro cytotoxicity and significant survival benefits in syngeneic AML mouse models, while showing manageable on-target/off-tumor toxicity in human VISTA transgenic mice.
