VISTA-knockout NK cells are an experimental adoptive cell therapy being investigated for the treatment of malignant gliomas, including glioblastoma multiforme (GBM). Developed by researchers at Nationwide Children's Hospital, these NK cells are modified using CRISPR/Cas9 ribonucleoprotein (RNP) editing to eliminate the expression of VISTA (V-domain Ig suppressor of T cell activation), a negative immune checkpoint also known as PD-1H. VISTA is highly expressed in the brain tumor microenvironment and normally functions to suppress immune cell activity. Preclinical studies indicate that VISTA-deficient NK cells possess superior tumor-killing capabilities, improved persistence, and a reduced susceptibility to exhaustion compared to wild-type NK cells, potentially offering a novel strategy to overcome the immunosuppressive barriers of solid tumors.
