Overview

Visugromab is a humanized, hinge-stabilized immunoglobulin G4 (IgG4) monoclonal antibody that targets and neutralizes growth/differentiation factor 15 (GDF-15), a key immunosuppressive cytokine produced by tumor cells. By blocking GDF-15, visugromab reverses immune suppression in the tumor microenvironment, restores T-cell infiltration and activation, and enhances anti-tumor immune responses. It has demonstrated the ability to overcome resistance to both checkpoint inhibitors (such as PD-1/PD-L1 antibodies) and antibody-drug conjugates (ADCs), leading to improved clinical outcomes in patients with advanced solid tumors who have relapsed or are refractory to prior immunotherapies. Additionally, visugromab shows potential for treating cancer-associated cachexia by inhibiting GDF-15-mediated activation of the GFRAL pathway in the brainstem, which drives weight loss and appetite suppression in cancer patients[1][3][4][5].
