Overview

VMP001 is a novel chimeric protein vaccine candidate developed to prevent infection and disease caused by *Plasmodium vivax*, the parasite responsible for vivax malaria. The vaccine incorporates the amino- and carboxy-terminal regions of the circumsporozoite protein (CSP) as well as a truncated repeat region containing sequences from both VK210 (type 1) and VK247 (type 2) strains of *P. vivax*. VMP001 is designed to induce immune responses targeting these CSP domains, aiming to provide broad protection against diverse *P. vivax* strains. In preclinical studies, VMP001 demonstrated immunogenicity in animal models, with protection associated with antibodies directed against the central repeat region of CSP[2]. In human phase 1/2a trials, it was shown to be safe and immunogenic but did not induce sterile protection; however, it delayed time to parasitemia in a significant proportion of vaccinated subjects[1]. The vaccine has been formulated with various adjuvants including GSK's AS01B system.
