Vixotrigine (also known as BIIB074 or CNV1014802) is an orally bioavailable, small molecule state-dependent sodium channel blocker that preferentially inhibits the voltage-gated sodium channel Nav 1.7 (SCN9A). Developed by Biogen following its 2015 acquisition of Convergence Pharmaceuticals, the drug is designed to treat various neuropathic pain conditions by selectively blocking sodium channels in a use-dependent manner. This mechanism is intended to provide potent analgesic effects while minimizing the central nervous system side effects and dose-titration requirements associated with non-selective sodium channel blockers like carbamazepine. Vixotrigine has been investigated in clinical trials for trigeminal neuralgia, lumbosacral radiculopathy (sciatica), and small fiber neuropathy, though several late-stage programs were discontinued following mixed clinical results.
