Overview

VK-2019 is a first-in-class, orally bioavailable small molecule inhibitor that selectively targets Epstein-Barr Nuclear Antigen 1 (EBNA1), a viral protein essential for the replication and maintenance of the Epstein-Barr virus (EBV) genome in latently infected cells. EBNA1 is consistently expressed in all known EBV-associated malignancies and acts as a master regulator of viral persistence and tumor cell proliferation. By inhibiting EBNA1, VK-2019 disrupts latent EBV replication, leading to reduced tumor growth in preclinical models of EBV-driven cancers such as nasopharyngeal carcinoma (NPC). The drug was invented by researchers at The Wistar Institute and has demonstrated an acceptable safety profile with on-target biological activity in early-phase clinical trials for advanced NPC. Clinical development is ongoing, with potential applications for other EBV-positive cancers including gastric carcinoma and lymphomas[4][5][6][7][8].
