VK-2735

VK-2735 is a novel dual agonist of the glucagon-like peptide 1 receptor (GLP-1R) and the glucose-dependent insulinotropic polypeptide receptor (GIPR), developed by Viking Therapeutics for the treatment of metabolic disorders, primarily obesity. As a dual incretin mimetic, VK-2735 activates both GLP-1 and GIP receptors to enhance insulin secretion in response to nutrient intake, suppress appetite, slow gastric emptying, reduce body weight, and improve insulin sensitivity. The drug is being developed in both oral tablet and subcutaneous injectable formulations. Clinical trials have demonstrated significant dose-dependent reductions in body weight with favorable safety and tolerability profiles; most adverse events are mild or moderate gastrointestinal symptoms. Phase 2 studies have shown up to 14.7% mean body weight reduction with subcutaneous administration over 13 weeks[1][2][3][4][8]. Viking Therapeutics plans to initiate Phase 3 development for the subcutaneous formulation in early 2025[1][4].
