Overview

**VL9-TCR-T** is an investigational T cell receptor-engineered T cell (TCR-T) therapy that exploits HLA-E-restricted T cell receptors targeting the VL9 peptide, a signal peptide-derived epitope cotranslationally released by signal peptide peptidase and loaded onto HLA-E/β2m dimers via TAP and Tapasin. Engineered T cells expressing these TCRs demonstrate potent and preferential cytotoxicity against VL9-presenting tumor cells (e.g., 84C cell line) compared to healthy PBMCs, with ~50% tumor cell killing versus ~30% PBMC viability in preclinical assays. Developed to leverage HLA-E presentation for broad applicability across HLA types, it represents a universal TCR-T approach for cancer immunotherapy, distinct from MHC-Ia-restricted therapies, with potential in hematologic and solid tumors.[1][3][5]
