VLPCOV-01 is a self-amplifying messenger RNA (sa-mRNA) vaccine candidate developed by VLP Therapeutics Japan for the prevention of COVID-19. Unlike conventional mRNA vaccines, sa-mRNA vaccines incorporate a replicase gene, typically derived from alphaviruses, which allows the mRNA to replicate itself once inside the host cell. This mechanism results in higher and more sustained levels of antigen expression, potentially allowing for lower dosing requirements compared to standard mRNA platforms. VLPCOV-01 specifically encodes the SARS-CoV-2 spike protein and is delivered via intramuscular injection. The vaccine entered clinical development in Japan to evaluate its safety, tolerability, and immunogenicity in healthy adults.
