VLPCOV-02 is a self-amplifying RNA (saRNA) vaccine developed as a booster for SARS-CoV-2 (COVID-19). It is encapsulated in lipid nanoparticles and encodes the receptor-binding domain (RBD) of the spike protein, specifically derived from the Brazil (gamma) variant of SARS-CoV-2. The vaccine incorporates 5-methylcytosine, a modified nucleoside base, to reduce innate immune responses and reactogenicity compared to earlier saRNA vaccines. Clinical studies have shown that VLPCOV-02 induces robust immunoglobulin G responses and has a favorable safety profile with mostly mild or moderate adverse events such as fatigue and myalgia. It is currently being evaluated in phase 1 and phase 3 clinical trials as a booster in individuals previously vaccinated against or infected with SARS-CoV-2[1][2][3][4][8].
