VLPCOV-05 is a self-amplifying mRNA (sa-mRNA) vaccine candidate developed by VLP Therapeutics Japan for the prevention of COVID-19. Utilizing a replicon platform, the vaccine encodes both the SARS-CoV-2 spike protein and a viral replicase complex. This technology allows the mRNA to replicate itself within the cytoplasm of host cells, leading to sustained and high-level expression of the target antigen. This mechanism potentially enables effective immunization at significantly lower doses compared to conventional mRNA vaccines. VLPCOV-05 is specifically being evaluated as a booster dose in adults previously vaccinated with mRNA vaccines. In Phase 3 clinical trials, it is being assessed for efficacy, safety, and immunogenicity against circulating SARS-CoV-2 variants, including comparisons with established vaccines such as COMIRNATY Omicron JN.1.
