Overview
**VLPONC-01 + pembrolizumab** is an investigational combination immunotherapy regimen currently in Phase 1 clinical trials for head and neck squamous cell carcinoma. VLPONC-01 is a virus replicon particle (VRP) encapsulated, self-amplifying RNA encoding interleukin-12 (IL-12), designed to be injected intratumorally to induce a potent anti-tumor immune response, particularly enhancing the tumor microenvironment. Pembrolizumab is a monoclonal antibody targeting programmed cell death protein 1 (PD-1), functioning as an immune checkpoint inhibitor, administered intravenously in this combination to block PD-1-mediated inhibition of T-cell immune responses. The combination aims to synergistically stimulate anti-tumor immunity, with VLPONC-01 boosting local immune activation through IL-12 and pembrolizumab relieving immune suppression by blocking the PD-1 pathway[2][4][7].
