Drug intelligence / Profile preview
VLR258 is an ActRIIB-based Fc ligand trap (recombinant fusion protein) being developed by Alaunos Therapeutics (following its acquisition of Velaris Therapeutics) for the treatment of pulmonary arterial hypertension (PAH) and related cardiopulmonary diseases, including secondary pulmonary hypertension associated with heart failure with preserved ejection fraction (HFpEF) and interstitial lung disease (ILD). Discovered by Paragon Labs, VLR258 is specifically engineered to target the ActRIIB (Activin Receptor Type IIB) signaling pathway while sparing BMP-9 and BMP-10, which are ligands associated with hematologic adverse events seen with other ActRII inhibitors. VLR258 is currently in preclinical development, with initial regulatory filings anticipated as a near-term milestone.
