VLS-1488 is a novel, orally bioavailable small molecule inhibitor of the mitotic kinesin protein KIF18A, developed for the treatment of advanced cancers characterized by chromosomal instability. By selectively binding to and inhibiting KIF18A, which is essential for proper chromosome alignment during mitosis in cancer cells but not required in normal diploid cells, VLS-1488 induces multipolar cell division and mitotic arrest. This leads to tumor cell death while sparing normal cells. Preclinical studies demonstrated dose-dependent inhibition of tumor growth in models with high chromosomal instability. In early clinical trials, VLS-1488 has shown promising anti-tumor activity—particularly in platinum-resistant high-grade serous ovarian cancer—and a favorable safety profile with no dose-limiting toxicities observed at clinically active doses[1][3][4][5][6][7].
