Overview

VLTR-557 is a long-acting anti-VEGF biologic developed by Valitor using its proprietary multivalent polymer (MVP) technology, which covalently conjugates multiple copies of a stability-optimized single-domain VHH antibody to hyaluronic acid for extended intravitreal durability. It exhibits high binding affinity to VEGF-A (Kd < 1 pM), rapid inhibition of VEGF signaling, and stability under physiological conditions for at least 6 months, with a vitreous half-life of 15.6 days in preclinical rabbit models—approximately 4 times longer than standard anti-VEGF therapies like aflibercept. Preclinical studies in rabbits and NHPs demonstrate sustained ocular tissue exposure, minimal systemic distribution (serum levels ~10,000x lower than vitreous), and good tolerability, supporting potential for >6 months of efficacy from a single intravitreal injection as a next-generation treatment for neovascular ocular diseases.[1][2][3][5][6]
