VMD-928 is an investigational, orally administered small molecule that acts as a selective antagonist of the tropomyosin receptor kinase A (TrkA), also known as the nerve growth factor receptor. By specifically binding to and inhibiting TrkA, VMD-928 blocks neurotrophin-TrkA interactions and prevents TrkA activation, which in turn inhibits downstream signaling pathways involved in tumor growth and pain transmission. Preclinical studies have shown that VMD-928 can decrease tumor growth rates and reduce pain in animal models. The drug is being developed primarily for the treatment of advanced solid tumors or lymphoma characterized by overexpression of TrkA protein, particularly in patients who have progressed on or are non-responsive to available therapies. Clinical trials have included patients with various cancers such as adenoid cystic carcinoma, cholangiocarcinoma, lung cancer, pancreatic cancer, parotid cancer, squamous cell carcinoma of head and neck, ovarian cancer, hepatocellular carcinoma, mesothelioma, thymic carcinoma among others[1][4][6][8].
