**VMT-01** is a targeted alpha-particle radiopharmaceutical designed to selectively bind melanocortin 1 receptor (MC1R), a protein overexpressed on metastatic melanoma tumors, and deliver the alpha-emitting isotope **212Pb** to induce potent DNA damage and tumor cell death via high linear energy transfer radiation. Developed by **Perspective Therapeutics**, it functions as a theranostic agent, using **203Pb-VMT-01** for pre-therapy imaging and patient selection via SPECT to confirm MC1R expression, followed by therapeutic **212Pb-VMT-01** dosing. It has received **FDA Fast Track Designation** for MC1R-positive unresectable or metastatic melanoma and is being evaluated in a phase 1/2a trial (NCT05655312) as monotherapy and in combination with nivolumab, showing promising early safety with no dose-limiting toxicities observed to date.[1][2][3][4][9]
