**vMyx-IL15Rα-tdTr** is a recombinant, genetically engineered myxoma virus (an oncolytic poxvirus) expressing a fusion protein of mouse interleukin-15 receptor alpha (IL15Rα) and interleukin-15 (IL15), along with the fluorescent reporter protein tdTomato red. This agent is designed to provide both direct oncolytic activity against tumors and potent enhancement of antitumor immune responses. The IL15Rα-IL15 fusion protein increases the stability and bioavailability of IL15 at tumor sites, leading to enhanced recruitment and activation of natural killer (NK) cells and CD8+ T cells in the tumor microenvironment. Preclinical studies demonstrate efficacy and improved survival in mouse models of aggressive melanoma and glioma, especially when combined with adoptive T cell therapy and prostaglandin synthesis inhibitors. The virus is non-pathogenic in non-rabbit hosts and shows tumor-specific infection and lysis, with a favorable safety profile compared to related oncolytic poxviruses[1][2][3][4][5][6][7].
