Overview

VNP40541 is a preclinical-stage, hypoxia-selective small molecule cytotoxin that was under development by Vion Pharmaceuticals. It was designed as a hypoxia-activated prodrug (HAP) to specifically target the oxygen-deprived regions of solid tumors, which are typically resistant to conventional chemotherapy and radiation. In the low-oxygen environment of a tumor, the compound is intended to undergo bioreductive activation, transforming into a potent cytotoxic agent (likely a DNA-damaging species) while remaining relatively inactive in well-oxygenated healthy tissues. Development of VNP40541 was halted when Vion Pharmaceuticals filed for Chapter 11 bankruptcy in 2009, and the asset has not significantly progressed in clinical development since that time.
